Delgocitinib Shows Real-World and Adolescent CHE Outcomes: Study

Key Takeaways

  • Interim REACHED data provide early real-world evidence for delgocitinib cream in adults with moderate to severe chronic hand eczema (CHE), with 28.6% assessed as clear or almost clear at Week 8.
  • Post hoc DELTA TEEN analyses found greater early reductions in patient-reported itch and pain with delgocitinib cream versus vehicle in adolescents aged 12 to 17 years.
  • At Week 16, 27.0% of adolescents receiving delgocitinib reported no CHE-related quality-of-life impact versus 0% with vehicle.

LEO Pharma presented new delgocitinib cream data at the 35th European Academy of Dermatology and Venereology (EADV) Congress, including interim real-world findings in adults and post hoc phase 3 analyses examining symptoms and quality of life in adolescents with moderate to severe chronic hand eczema (CHE).

DELTA TEEN and REACHED Data Examine Delgocitinib Across Age Groups

A post hoc analysis of the phase 3 DELTA TEEN trial evaluated daily Hand Eczema Symptom Diary itch and pain scores among adolescents aged 12 to 17 years. Mean reductions from baseline on Day 1 were greater with delgocitinib cream than vehicle for itch (–1.03 vs –0.20; P = 0.040) and pain (–1.34 vs –0.39; P = 0.019), with differences also reported across the first 2 weeks.

A separate post hoc analysis evaluated health-related quality of life using the Children’s Dermatology Life Quality Index (CDLQI). At Week 16, 27.0% of adolescents receiving delgocitinib had a CDLQI score of 0, indicating no reported impact on quality of life, compared with 0% receiving vehicle (P = 0.005). The proportion reporting a large or very large impact decreased from 21.6% to 4.1% with delgocitinib and from 25.0% to 16.7% with vehicle.

Meanwhile, interim findings from REACHED, an ongoing prospective, noninterventional German study, provide early real-world data in adults. Among patients with available Week 8 assessments, 28.6% were rated clear or almost clear on the modified Physician’s Global Assessment, compared with none at baseline. Mean Hand Eczema Severity Index scores decreased from 62.4 to 30.8, and 35.5% achieved at least 75% improvement.

All reported adverse events in the interim REACHED analysis were nonserious, with all but 1 classified as mild or moderate. The findings remain limited by the interim, noncomparative nature of the real-world analysis and the post hoc design of the adolescent analyses.
