New LATITUDE Data Detail Zasocitinib Efficacy, Durability in Plaque Psoriasis
CONTENT
Key Takeaways
- In the Phase 3 LATITUDE Atlas head-to-head trial, zasocitinib produced higher week 16 PASI 100, PASI 90, and sPGA 0 response rates than deucravacitinib in adults with moderate-to-severe plaque psoriasis.
- Additional LATITUDE PsO 3001 and 3002 analyses showed maintenance of skin clearance among week 24 responders and early improvements in patient-reported itch and quality of life.
- Zasocitinib remains investigational and is currently under regulatory review in the United States and European Union.
New Phase 3 data presented at the European Academy of Dermatology & Venereology (EADV) Congress 2026 showed higher skin clearance rates with investigational oral tyrosine kinase 2 (TYK2) inhibitor zasocitinib (TAK-279) compared with deucravacitinib in adults with moderate-to-severe plaque psoriasis.
In the randomized, multicenter, double-blind Phase 3 LATITUDE Atlas study, 606 patients received zasocitinib 30 mg once daily or deucravacitinib 6 mg once daily through week 16. The primary endpoint was achievement of 100% improvement in Psoriasis Area and Severity Index (PASI 100) at week 16.
Zasocitinib Achieves Higher Week 16 Skin Clearance Rates
At week 16, PASI 100 was achieved by 36.5% of patients receiving zasocitinib (n = 301) compared with 13.9% receiving deucravacitinib (n = 303; P < .001). PASI 90 responses were reported in 62.5% and 34.0% of patients, respectively (P < .001), while static Physician Global Assessment (sPGA) 0 responses occurred in 43.2% and 18.5%, respectively (P < .001). Zasocitinib met the primary and key secondary efficacy endpoints versus deucravacitinib.
Separate analyses of the Phase 3 LATITUDE PsO 3001 and 3002 trials evaluated longer-term efficacy and patient-reported outcomes. Among patients who responded at week 24, up to 93% maintained skin clearance through week 52 in PsO 3001 in a post hoc analysis and through week 40 in PsO 3002. Improvements in itch were reported as early as week 2, and Dermatology Life Quality Index 0/1 responses improved by week 4 compared with placebo and, in PsO 3002, apremilast; these comparisons used nominal P values.
No new safety signals were identified. The most commonly reported adverse events across the Phase 3 LATITUDE psoriasis program included upper respiratory tract infection, nasopharyngitis, and acne.
