Rubedo Life Sciences announced plans to evaluate its investigational GPX4 modulator RLS-1496 in patients with androgenetic alopecia (AGA), following ex vivo findings showing increased growth of human hair follicles exposed to the agent, according to an announcement from the manufacturer.

The clinical study is expected to begin in October 2026, with initial results anticipated during the first half of 2027. David Pariser, MD, FACP, FAAD, professor of dermatology at Eastern Virginia Medical School and principal investigator at Virginia Clinical Research, Inc., will lead the trial.

RLS-1496 Targets Senescent Cells in Androgenetic Alopecia

RLS-1496 is being developed as a SenoAdaptive drug designed to selectively eliminate or restore function in senescent and stressed cells. According to Rubedo, the approach targets cellular processes that contribute to hair follicle miniaturization and impaired regeneration in AGA.

In an independent ex vivo model, intact hair follicles obtained from male patients with AGA were challenged with dihydrotestosterone to model features of AGA. RLS-1496 increased follicle elongation compared with vehicle at the tested doses. The highest concentration produced numerically greater mean elongation than finasteride, although the company did not provide the numerical results, sample size, concentrations, or statistical comparisons with finasteride in the announcement.

Rubedo said the findings were consistent with rescue of the anagen growth phase and reactivation of follicular stem cells. The company also reported that its spatial transcriptomic analyses identified senescence markers and signals associated with follicular regeneration. The ex vivo findings are preclinical and do not establish efficacy or safety for hair loss in humans, the manufacturer noted. 

“The preliminary data we have seen with RLS-1496 are scientifically intriguing and warranted clinical evaluation,” Pariser said. “The senescence biology underlying AGA is increasingly well-supported in the published literature, and RLS-1496 represents a genuinely novel approach to it.”
