SYX-5219 Improves Eczema Severity, Itch in Early Clinical Trial

Key Takeaways

  • SYX-5219, an investigational oral pyruvate kinase M2 (PKM2) modulator, demonstrated improvements in atopic dermatitis severity and itch in a Phase 1b trial.

  • At 6 weeks, placebo-adjusted differences were approximately 28% for EASI and 48% for PP-NRS, according to Sitryx Therapeutics.

  • The company plans to initiate Phase 2 trials in the first half of 2027.

An investigational oral therapy targeting immune cell metabolism demonstrated improvements in disease severity and itch among adults with moderate to severe atopic dermatitis (AD), according to Phase 1b results announced by Sitryx Therapeutics.

SYX-5219, a first-in-class pyruvate kinase M2 (PKM2) modulator, met the company's exploratory efficacy targets and was reportedly well tolerated during 6 weeks of treatment. The company announced the results in a press release.

Phase 1b Atopic Dermatitis Trial Results

The randomized, double-blind, placebo-controlled trial included 35 adults with moderate to severe AD across clinical sites in the United States and Europe. Participants received SYX-5219 or placebo in a 2:1 ratio for 6 weeks, followed by a 14-day off-treatment observation period.

According to Sitryx, treatment differences emerged by day 8 and increased throughout the dosing period. At day 43, the placebo-adjusted difference in Eczema Area and Severity Index (EASI) score was approximately 28%, while the difference in Peak Pruritus Numerical Rating Scale (PP-NRS) itch score was approximately 48%.

Investigators also reported improvements in validated Investigator Global Assessment (vIGA) scores. Biomarker analyses identified correlations between improvements in disease activity and reductions in CCL17/TARC and CCL11/eotaxin.

"SYX-5219's novel PKM2 approach and these encouraging early results make it an important candidate for further investigation," said trial principal investigator Alan Irvine, MD, DSc, professor of dermatology at Trinity College Dublin.

The findings remain preliminary, with detailed safety data and statistical analyses pending. Sitryx anticipates initiating Phase 2 development in the first half of 2027.
